  • IMPLANTATION OF ALLOGENIC OSTEOBLAST COMBINED WITH CALCIUM PHOSPHTA COMPOSITES

    The aim of this experiment was to study the osteogenesis in vivo of allogenic osteoblast combined culture with calcium phosphate composites. The osteoblasts were obtained by enzymatic digestion of periosteum from fibula subcultured to 13 generations, the cells were combined culture with hydroxyapatite and biphasic calcium phosphate. Subseguently, the composite was implanted into rabbits subcutaneously or intramuscularly. The blank material was implanted in the contralateral side as control. Four weeks later, all animals were sacrificed. All the implants were examined by gross observation, histological examination and EDXA. The results showed: 1. obvious ingrowth of connective tissue with very little inflammatory reaction; 2. new bone formation in the composites with deposit of Ca and P on the surface of osteoblast, but none in the blank materials; 3. no significant difference of new bone formation between the different sites of implantation or different materials, but those implanted intramuscularly had lamellae form of new bone while those implanted subcutaneously had only mineralization of extracellular matrix. The conclusion were: 1. the composites are biocompatible with prior osteogenesis property; 2. periosteal-derived allogenic osteoblasts obatined by enzymatic digestion could survive following implantation with bioactivity; 3. rich blood supply might be advantageous to new bone formation and its maturation.

  • EXPERIMENTAL STUDY OF INSTANTANEOUS EXPANSION OF SOFT TISSUE

    Abstract An animal experiment had been conducted for the purpose to find out the possibility of application of instantaneous sustained limited expansion (ISLE). A total of 54 skin defects in 9 pigs were obtained and were divided into two groups. One group of the pigs were subjected to instantaneous sustained limited expansion, the others, as the control group, just received the tension suture. At the same time the blood supply of the skin was measured by laser doppler flowmeter (LDF). It was showed that microcirculatory perfusion of the ISLE group was considerably better than that of the control. Inaddition, the correlation between the blood flow of the skin and the expanding pressure, as well as the pathologic changes of the skin were discussed.

  • EXPERIMENTAL STUDY OF ANASTOMOSIS OF PERIPHERAL NERVE WITH BIOLOGICAL ADHESIVE AGENT(PRELIMINARY REPORT)

    In this experiment ,the sciatic nerves or twen-ty-eight rats were cut and then anastomosed withbiological adhesive agent or by suture in randonand the latter as control. The biological adhesive a-gent from human plasma was composed of fibrino-gen coagulase and medullary sheath of the nervetaken at the anasomosis region were studied histol-gically.   • EXPERIMENTAL STUDY ON PERMEABILITY OF SKIN EXPANDERS TO METRONIDAZOLE AND PROCAINE

    An in vitro experiment showed that the skin expanders were permeable to metronidazole and procaine. Twenty kidney shaped skin expanders were divided into four groups. Group 1. 100ml 0.2% metronidazole solution was injected into the expanders and the expenders were immersed in a flask filled with 100ml saline solution, and then were placed in a hermetically sealed glass chamber; Group 2.the whole procedure was the same as that of Group 1 except the expander was previously boiled in water for 30 minutes; Group 3. 100 ml 2% procaine was injected instead of metronidazole, other step was the same as that ofgroup I; and Group 4. the whole procedure was the same as that of Group 2 except the solutioninjected was 2% procaine. The concentration of metronidazole and procaine in the surrounding saline was measured at 1st, 2nd, 4th, 16th, 24th, 48th, 72nd 120th hours. The rate of diffusion of a drug was highest at 2 and 4 hous. The rate of diffusion was inversely proportional to its molecular weight, i.e., the smaller the molecular weight the greater the permeability. In view of this, during the process of expansion, metronidazole and procaine would diffuse out of the expander which might be beneficial for preventing infection and controlling pain.

  • EXPERIMENTAL STUDY OF WHOLE-LAYER AND PART-LAYER ANASTOMOSIS OF SMALL ARTERY

    Using 70 SD white rats, diveded in two groups at random, after the common carotid artery wa(?) exposed, anastomosis of the artery was done by whole-layer suture and suture without including the endothelial layer, respectively. The rate of patency of both groups immediately after operation was 100 percent, where as in late stage, 94 percent and 97 percent, respectively. From the histologic exam ination, it was found that in the group of whole-layer suture, the time required to cover the sutureline with endothelium was delayed and there was rupture of the clastic fibers.

  • SEQUENTIAL EXPERIMENTAL STUDY OF DENERVATED NEURO-MUSCULAR JUNCTIO

    Motor endplate is the structure connected between the nerve terminal and muscle fibre and plays a very important role in conducting nerve impulses to the target, therefore, systemical study of the sequential changes of the motor end-plate upon denervation is quite important.Ninety New Zealand rabbits were divided into nine groups from two weeks to nine months after denervation. Acetycholinesterase(AchE) was analyzed quantitatively to study the sequential changes of the motorendplates of tibialis anterior muscle. The results showed that there was no significant reduction of AchE at theend if one month after denervation, whereas a sharp reduction of AchE afterwards. AchE could not be stained after five month denervation.

  • EXPERIMENTAL STUDY OF EFFECT OF MENISCUSSUTURE ON MENISCUSHEAL ING

    OBJECTIVE To investigate the effect of meniscus suture on meniscus healing which included healing time and healing pattern. METHODS Fourty healthy rabbits were adopted in this study. The model of meniscus injury was made by a longitudinal incision at the medial meniscus of the left knee. The rabbits were divided into two groups, the experimental group was treated by meniscus suture and the control group was unsutured. After operation, the meniscus samples were collected periodically and observed by gross, light and electronic microscope to analysis the meniscus healing. RESULTS The injured meniscus was healed gradually and completely at the sixth week in the experimental groups. More fibroblasts and less fibrocartilage cells could be observed in the healed meniscus. Oppositely, there was no meniscus healing in the control group and the edge of injured meniscus was sealed by epithelioid cells. CONCLUSION The meniscus suture can accelerate the healing process of meniscus injury. Besides, early suture make the injured meniscus correctly positioned to ensure the normal healing process.

  • EXPERIMENTAL STUDY ON POROUS HYDROXYAPATITE CERAMICS IN REPAIR OF SKULL BONE DEFECT OF RABBIT

    In order to investigate the possibility of porous hydroxyapatite ceramics (HAC) in the repair of skull bone defect, twenty-four rabbits were used. The bone defect model was created by operation to obtain a defect in parietal bone in a size of 1 cm x 1 cm. Filled the defect with HAC and methyl-methacrylate-syrene copolymer (MMAS) to fill the defect as control. At 1st, 2nd and 3rd months after operation, behavior of the rabbits was observed and then these animals were sacrificed and specimens were examined under microscope. Results showed as follows: after operation, behavior of all animals were normal. By histological examination, it was found that in HAC group, there were granulation tissue, fibrous tissue and newly formed vessels grew into the pores and the osteoblasts formed osseous trabeculae. There was no inflammatory cell infiltration. In the MMAS grafted asea, there was formation of fibrous membrane. It suggested that HAC might be a good material for bone substitute in repair of skull bone defect.

  • Experimental study on the in vivo continuous measurement of pressure difference between the anterior and the posterior chambers

    A set of device for the in vivo measurement of the pressure difference between the anterior and the posterior chambers (PDAP) was designed to investigate the temporal varying rules of PDAP in the anterior segment of rabbit eyes. A platform was established for the in vivo measurement of PDPA according to the mechanism of joint implement. Rabbit models with high intraocular pressure (IOP) were constructed by means of injecting Carbomer into anterior chamber to increase IOP. The in vivo 24 hours continuous measurements of PDAP were performed for normal rabbit eye and eye with high IOP. The developed device could sensitively response to the small pressure difference in eye. The pressure difference in the normal rabbit eye varied with time, and the variation range during a whole day was 5.84–96.84 Pa which reflected the existence of physiological rule. For the rabbit eye with high IOP, pressure in anterior chamber was higher than that in posterior chamber which was in consistence with the theory of self-adaptation adjustment. The present study indicates that the approaches and device designed in this paper can well implement the measurement of PDAP as well as the temporal varying rules of PDAP in the anterior segment during a whole day.
