  • DYNAMIC MEASUREMENT AND SIGNIFICANCE OF ENDOTHELIN IN BILE AND PERIPHERAL VEINOUS BLOOD IN PATIENTS WITH ACUTE CHOLANGITIS OF SEVERE TYPE

    Objective To evaluate the relationship between endothelin (ET) in bile and peripheral blood with systemic and hepatobiliary injury in patients with acute cholangitis of severe type (ACST). Methods ET, ALT and total bilirubin in bile and peripheral veinous blood of 25 patients with acute cholangitis of severe type (ACST) were detected during operation, one week and two weeks after operation. Results The contents of ET, ALT and total bilirubin were significantly lower on 7-day and 14-day after operations as compared with that during operations (P<0.05 and P<0.01). The concentration of ET in peripheral veinous blood paralleled with that in bile. Conclusion This suggests that ET is tightly related with the pathologic process of ACST. So, in patients with ACST, the dynamic measurement of ET in peripheral veinous blood can be an index for judging the degree of pathological damage either to the hepatobiliary or systemic systems.

  • Effects of BQ123 on Carcinoma Angiogenesis of Implanted Laryngeal Carcinoma

    摘要:目的: 探讨选择性内皮素A受体拮抗剂BQ123对人喉癌Hep2细胞裸鼠种植瘤的生长及血管形成的影响。 方法 :将实验动物裸鼠随机分为3组:BQ123[n =8,2mg/(kg·day)]、氟尿嘧啶组[n =8,2mg/(kg·day)]、生理盐水组(n =8),比较各组裸鼠成瘤体积、微血管密度(MVD)。 结果 :BQ123组肿瘤体积为(162±053)cm3,明显小于生理盐水组及氟尿嘧啶组,差异具有统计学意义;BQ123组的肿瘤组织中MVD高倍镜下为232,明显低于生理盐水组(586)及氟尿嘧啶组(395),差异具有统计学意义。 结论 :BQ123对人喉癌Hep2细胞在裸鼠体内有明显抑瘤作用,肿瘤的体积、肿瘤组织MVD显著低于对照组,表明BQ123可通过抑制肿瘤血管生成而显著抑制肿瘤生长。Abstract: Objective: To study the effects of endothelin A receptor blockade BQ123 on the implanted human laryngeal carcinoma angiogenesis of nude mouse. Methods : From March 2008 to July 2009, 24 Balb/c nude mice were randomly divided into three groups: BQ123 group [〖WTBX〗n =8, BQ123 at 2mg/(kg·day)], 5Fu group [〖WTBX〗n =8, fluorouracil at 2mg/(kg·day)] and the control group (〖WTBX〗n =8, normal saline). The carcinoma volume and microvascular density of each group were compared. Results : The tumor size of BQ123 group, which was (162±053)cm3 in average, was significant smaller than the tumor sizes of the other two group s. The average microvascular density score of the tumors in BQ123 group was 232 per hyper power len (HP), which was also significantly less than the average scores of control groups (586 and 395 respectively). Conclusion : Nude mouse experiments show that the carcinoma volume and microvascular density of BQ123 group are significantly lower than those of the control groups. BQ123 inhibits the growth of carcinoma by its inhibition of carcinoma angiogenesis.

  • Achievements of Basic Research in Small-for-Size Liver Transplantation

    Objective To introduce the mechanisms of graft injuries after small-for-size liver transplantation and protective measures. Methods Recently relevant literatures were reviewed and summarized. Results Portal hypertension after small-for-size liver transplantation induces mechanical injuries as well as hepatic sinusoidal microcirculation disturbance and cytokines release, which worsened the injuries. Decrease portal pressure by surgery or drug could improve grafts function. ConclusionComprehending the mechanisms of graft injuries will contribute a lot for the living donor liver transplantation.

  • The correlation of serum 8-hydroxydeoxyguanosine and endothelin-1 levels with cognitive dysfunction in COPD patients

    ObjectiveTo investigate the correlation and clinical significance of 8-hydroxydeoxyguanosine (8-OHdG) and endothelin-1 (ET-1) levels with cognitive dysfunction in patients with chronic obstructive pulmonary disease (COPD), and provide new idea for the prevention and treatment for cognitive dysfunction in COPD patients.MethodsA total of 103 COPD patients, according to the Montreal cognitive assessment scale standard for evaluation, were divided into a cognitive dysfunction group and a cognitive normal group. Serum 8-OHdG and ET-1 levels were compared between the two groups and their correlations with cognitive function were analyzed with the receiver operating characteristic (ROC) curve.ResultsThe levels of serum 8-OHdG and ET-1 in the COPD patients with cognitive impairment were significantly higher than those in the cognitive normal group [8-OHdG: (13.91±9.04) ng/ml vs. (7.28±3.00) ng/ml; ET-1: (95.64±57.66)pg/ml vs. (69.20±7.89)pg/ml] (both P<0.05). The levels of 8-OHdG (OR=22.94, 95%CI 7.06-74.53) and ET-1 (OR=19.76, 95%CI 6.59-59.31) were associated with cognitive impairment in the COPD patients. The areas under ROC curve of serum 8-OHdG and ET-1 levels to predict cognitive dysfunction in the COPD patients were 0.786 (95%CI 0.691-0.881) and 0.790(95%CI 0.695-0.885).ConclusionsThe serum levels of 8-OHdG and ET-1 are associated with cognitive impairment in COPD patients. The levels of 8-OHdG and ET-1 in serum can predict cognitive impairment with high specificity.

  • Protective Effect of Non-peptide Endothelin Antagonist Ferulic Acid on Lung Injury During Cardiopulmonary Bypass.

    Objective To investigate the role of endothelin(ET) in lung injury during cardiopulmonary bypass (CPB) and study the possible mechanism of ET-mediated lung injury and the protective effect of ferulic acid(FA) during the procedure. Methods Twelve dogs were randomly divided into 2 groups and models of CPB with pulmonary perfusion were established by perfusion of 4 C FA solution through proximal pulmonary artery in the experiment group while control group only received 4 C crystal cardiac arrest solution without pulmonary perfusion. Changes in the content of ET, NO, malonaldehyde (MDA), dry to wet (D/W) in lung tissue and lung function- related indices PaO2/FiO2, airway pressure (AWP), pulmonary vascular resistance (PVR), lung compliance before and after CPB in both groups were measured respectively. Results ET content increased after CPB in control group (P〈0. 05) ,while experiment group had a lower level of ET than that of control group (P〈0.05); D/W, MDA levels in experiment group decreased (P〈0. 05), but NO content increased (P〈0. 05) as compared with control group. After pulmonary perfusion with FA, PaO2/FiO2 and lung compliance values in experiment group were higher than those of control group (P〈0.05),AWP, PVR values lowered accordingly(P〈0. 05). Lung injury was less severe in the experiment group. Conclusion ET is involved in pathogenesis of lung injury during CPB, FA can effectively reduce lung injury and improve lung function thus having a good protective effect on the lung.

  • 室间隔缺损伴肺动脉高压患者血浆内皮素、一氧化氮与肺细小动脉形态变化的关系

    目的 探讨内皮素(ET-1)、一氧化氮(NO)在肺动脉高压(PH)发生、发展过程中的作用. 方法 60例室间隔缺损患者分为两组,无PH组:20例,不合并PH,平均肺动脉压(MPAP)lt;20mmHg(1kPa=7.5 mmHg);PH组:40例,合并PH,MPAPgt;20mmHg.测定两组血浆ET-1和NO含量,作肺组织活检,观察肺血管的病理改变及测定肺细小动脉显微形态指标. 结果 全部肺标本按阮英茆的Ⅳ级分类进行病理分级:无PH组患者肺血管无明显病理改变,PH组患者Ⅰ级7例、Ⅱ级13例、Ⅲ级20例,未见Ⅳ级病理改变.随着肺血管病理改变分级的增加,血浆ET-1、NO、血流动力学指标及肺细小动脉显微形态指标测定均呈阶梯样变化,除NO、血管腔面积/血管总面积(EA/TA)逐渐减小,其它指标均逐渐增高,ET-1、NO、MPAP、肺血管阻力(PVR)、肺细小动脉中膜厚度(mMTPA)、血管壁面积/血管总面积(WA/TA)、EA/TA和肌型肺细小动脉/微动脉总数的比值(TWPV),无PH组与PH组Ⅰ级、PH组Ⅰ级与Ⅱ级、Ⅱ级与Ⅲ级之间差别均有显著性意义(Plt;0.05,0.01);血浆ET-1与MPAP、PVR、mMTPA、WA/TA和TWPV均呈显著正相关,血浆NO均呈显著负相关;血浆ET-1与EA/TA呈显著负相关,血浆NO呈显著正相关(Plt;0.01);血浆ET-1浓度与血浆NO浓度呈显著负相关(r=-0.3452,P=0.0069). 结论 ET-1增高,NO减少,ET-1/NO动态平衡的破坏,参与了PH的发生、发展过程.

    To investigate the effect of modified cold preservation on rat livers. The modified method was that the vascular bed of rat liver was infused with an additional 20 ml, 30 ml and 40 ml self-made HYD solution/100g liver. Via portal vein and the vesselses of liver were ligated. Forty rats were randomly divided into four groups including conventional storage group (control group) and modified preservation groups (20%, 30% and 40% group). The indices of hepatic microcirculation, enzymes level in the effluent and bile production were determined in the groups. Results: Bile production and all the indices of hepatic microcirculation including portal perfused pressure, endothelin-1 in the effluent, Trypan Blue distrubution time and histology in experimental groups were significantly superior to those in control group (P<0.05). The liver enzymes in 30% group were markedly lower than those in control group (P<0.05). The preservation effect of rat livers in 30% group was the best among the experimental groups. Conclusions: The modified cold storage method is effective and may have potential for clinical application for liver preservation.

  • Experimental Study on Effects of Endothelin and Xuesaitong Injection on Hepatic, Renal and Myocardial Tissues in Obstructive Jaundice

    【Abstract】ObjectiveTo study the effects of endothelin (ET) and Xuesaitong injection on hepatic, renal and myocardial tissues after bile duct ligation (BDL) in rabbits. MethodsSeventytwo rabbits were randomly divided into three groups: BDL group (24 rabbits), BDL+Xuesaitong injection group (24 rabbits), and sham operation group (24 rabbits). Each group was subdivided into four subgroups of postoperative 3, 6, 9 and 12 d (6 rabbits in each subgroup). Automatic biochemical analysis equipment was used to detect the levels of serum TBIL, ALT, BUN and Crea. The levels of ET in plasma, hepatic, renal and myocardial tissues were measured with radioimmunological method. ResultsThe levels of ET in plasma, hepatic, renal and myocardial tissues in both BDL group and BDL+Xuesaitong injection group were higher than those of sham operation group (P<0.01). The levels of ET in plasma and tissues of BDL+Xuesaitong group were lower than those in BDL group (P<0.05). ConclusionObstructive jaundice can lead to an increase of ET in plasma, hepatic, renal and myocardial tissues, the level of ET increases with the time of obstruction. Xuesaitong injection may play a protective role in the injury of hepatic, renal and myocardial tissues after obstruction by decreasing the level of ET in plasma and tissues.

  • THE ROLE OF ENDOTHELIN IN THE PATHOGENIC MECHANISM OF ACUTE PANCREATITIS IN RATS

    Objective To study the effects of endothelin (ET) on acute pancreatitis in rats. Methods The acute edematous pancreatitis (AEP) and hemorrhagic necrotizing pancreatitis (AHNP) model of rats were induced by cerulein and dextran-110 and endothelin-1 was administered on AEP rats via intravenous injection. Serum amylase, plasma ET and 6-Keto-PGF1α, pancreas hostologic observation were determined. Results The serum concentration of amylase increased markedly in AHNP rats, and there was a significant difference between AHNP and AEP (P<0.01). A dose of extrinsic ET-1 may induce the conversion of AEP to AHNP in rats. The degree of pancreatic damage correlates positively with the level of the plasma ET. Conclusion Endothelin might take part in the development of acute pancreatitis, and play a key role in the conversion of AEP to AHNP.
