  • Changes of Fibrogenic Cytokines in Acute Paraquat Poisoned Rats and Effects of Pyrrolidine Dithiocarbamate

    Objective To investigate the mechanismof lung injury caused by paraquat poisoning by observing the changes of fibrogenic cytokines in acute paraquat poisoned rats and the effects of pyrrolidine dithiocarbamate ( PDTC) . Methods Sprague-Dawley rats were randomly divided into three groups, ie. acontrol group ( n =6) , a PDTC group ( n =36) , a paraquat group ( n = 36) , and a paraquat + PDTC group( n =36) . The rats in the PDTC group, the paraquat group, and the paraquat + PDTC group were subdivided into 6 subgroups sacrificed respectively on 1st, 3rd,7th,14th, 28th and 56th day after the treatment. The levels of transforming growth factor-β1( TGF-β1 ) , platelet-derived growth factor ( PDGF) , insulin-like growthfactor-1 ( IGF-1) in serum were measured. Meanwhile the expression of connective tissue growth factor ( CTGF) and hydroxyproline in lung tissues were detected. The relationship of above cytokines with hydroxyproline was analyzed. Results The destructive phase in early ( 1 ~7 d) was characterized by hemorrhage, alveolar edema, and inflammatory cell infiltration. The proliferous phase in later stage ( 14 ~56 d) was characterized by diffused alveolar collapse with fibroblast proliferation and patchy distribution of collagen fibers. Compared with the control group, the level of TGF-β1 on all time points, the level of PDGF from7th to 56th day, the level of IGF-1 from3rd to 56th day in the paraquat group all significantly increased ( P lt;0. 01) . Immunohistochemistry results showed CTGF positive cells mainly located in aleolar epithelialcells, endothelial cells,macrophages in early stage, and fibroblasts were main positive cells on the 28th and the 56th day. The expression of CTGF in the paraquat group increased gradually compared with the control group on different time points ( P lt; 0. 05 or P lt; 0. 01) . Meanwhile, the levels of above cytokines were positively correlated with the level of hydroxyproline. Noteworthy, PDTC treatment led to significant decreases of above cytokines compared with the paraquat group in corresponding time points ( P lt;0. 05 or P lt;0. 01) .Conclusions Over expressions of IGF-1, TGF-β1 , PDGF, IGF-1 and CTGF may play important roles in lung fibrosis of paraquat poisoned rats. PDTC, as a b NF-κB inhibitor, may inhibits NF-κB activity and further significantly decreases expressions of cytokines, leading to significantly attenuated pulmonary inflammation and fibrosis. However, the mechanisms of PDTC intervention still remain to be explored.

  • Biomechanical properties of epithelial mesenchymal transition in idiopathic pulmonary fibrosis

    Idiopathic pulmonary fibrosis (IPF) is a progressive scar-forming disease with a high mortality rate that has received widespread attention. Epithelial mesenchymal transition (EMT) is an important part of the pulmonary fibrosis process, and changes in the biomechanical properties of lung tissue have an important impact on it. In this paper, we summarize the changes in the biomechanical microenvironment of lung tissue in IPF-EMT in recent years, and provide a systematic review on the effects of alterations in the mechanical microenvironment in pulmonary fibrosis on the process of EMT, the effects of mechanical factors on the behavior of alveolar epithelial cells in EMT and the biomechanical signaling in EMT, in order to provide new references for the research on the prevention and treatment of IPF.

  • The Clinical Implication of Serum CA19-9 in Patients with Pulmonary Interstitial Fibrosis

    ObjectiveTo investigate the implication of serum CA19-9 in patients with pulmonary interstitial fibrosis (PF). MethodsOne PF patient was followed up for 5 years, and the levels of serum CA19-9, arterial oxygen pressure and changes of chest CT scanning were recorded and analyzed.Furthermore, another 24 PF patients with the level of serum CA19-9 exceeding the cut-off value (37 U/mL) and 30 healthy controls were recruited, and the pulmonary function was compared between the PF patients and the healthy controls. ResultsDuring the 5-year follow-up, the serum CA19-9 increased gradually from 53.45 U/mL to > 1200 U/mL with progressive reticulation of the lungs proven by the chest CT scanning, and the arterial oxygen pressure in rest without oxygen inhalation decreased from 54 mm Hg to 32 mm Hg.Compared to the healthy controls, the PF patients had significantly higher serum CA19-9 levels [73.8(38.1-1200.0) U/mL vs.6.9(2.0-19.1) U/mL, P < 0.01] and worse diffusion function [55.0(12.7-87.8) mL·min-1·mm Hg-1 vs. 90.4(82.0-134.0) mL·min-1·mm Hg-1, P < 0.05].There was a significantly negative relationship between the elevation of CA19-9 and diffusion function (r=-0.997, P < 0.01). ConclusionSerum CA19-9 may increase in PF patients, which may be an indicator for poor prognosis.

  • Protective Effects of Curcumin on Oxidative Stress-Induced Cell Injury in Human Fetal Lung Fibroblasts Co-cultured with A549 Cells and Its Mechanism

    Objective To investigate the effects of curcumin on oxidative stress in the co-culture system including human fetal lung fibroblasts and A549 cells, and discuss the potential and protective mechanism of the prophylactic effect of curcumin on pulmonary fibrosis. Methods The human fetal lung fibroblasts co-cultured with A549 cells were divided into five groups. The cells in the control group were cultured in DMEM without TGF-β1 or curcumin. The cells in the TGF-β1 group were cultured in DMEM containing 5 ng/mL TGF-β1 . In three TGF-β1 + cucurmin treatment groups, the cells were cultured in DMEM containing 5 ng/mL TGF-β1 and three different concentration of curcumin( 5, 10, 20 μmol /L, respectively) . ELISA was used to analyze the content of TNF-α. Serum level of MDA and SOD were tested by spectrophotometric analysis. Intracellular ROS production was detected by flow cytometry. NF-κB was measured by western blot. Results The serum MDA, intracellular ROS, the content of TNF-αand NF-κB protein expression in the TGF-β1 group were significantly increased while the activity of SOD was significantly decreased( P lt; 0. 01) , suggesting that the oxidative level of human fetal lung fibroblasts was obviously increased after TGF-β1 stimulation. After intervening by different concentration of curcumin, the serum MDA, intracellular ROS, content of TNF-αand NF-κB were significantly decreased while the activity of SOD was obviously increased( P lt;0.01) . Conclusion Low concentration of curcumin can reduce the oxidative level of human fetal lung fibroblasts co-cultured with A549 after TGF-β1 stimulation, and significantly increase the level of SOD, implying that curcumin may intervene pulmonary fibrosis by reduce oxidative level.

  • Effect of curcumin on bleomycin-induced pulmonary fibrosis in rats

    Objective To study the inhibitory effects of curcumin on bleomycin-induced pulmonary fibrosis in rats at the fibrosing stage and explore its possible mechanism.Methods 96 male SD rats were randomly divided into a normal control group,a fibrosis model group,a fibrosis model treated with prednisone group and a fibrosis model treated with curcumin group.Pulmonary fibrosis were induced by instilled bleomycin through tracheal.From day 15 after bleomycin administration,the curcumin group and prednisone group were given curcumin(300 mg/kg) or prednisone(5 mg/kg) per day by intragastric administration,respectively.The normal control group and fibrosis model group were given 1% sodium carboxymethyl cellulose(10 mL/kg) as control.Six rats of each group were randomly sacrificed on day 21,28,42 and 56 after bleomycin administration,respectively.The histological changes of the lung were evaluated by HE and Masson’s trichrome staining.Lung expressions of transforming growth factor-β1(TGF-β1) and hydroxyproline were assessed by immuno-histochemistry and digestion method,respectively.Results Pulmonary fibrosis and hydroxyproline level in the curcumin group were significantly reduced as compared with those in the model group on day 42 and 56.The expession of TGF-β1 in the curcumin group was significantly lower than that in the model group on day 28,42 and 56,and was not significantly different from the normal group on day 56.Conclusion Curcumin could alleviate bleomycin-induced pulmonary fibrosis in rats at the fibrosing stage by inhibiting the expressions of TGF-β1.

  • A pilot study of acid inhalation-induced pulmonary fibrosis in rats

    Objective To study the pathology and possible mechanism of experimental hydrochloric acid(HCl) inhalation-indued pulmonary fibrosis in rats.Methods 120 male SD rats were randomly divided into a nomal control group,a bleomycin group,a high dose HCl group,a middle dose HCl group and a low dose HCl group.The bleomycin group was intratracheally injected with bleomycin once to induce pulmonary fibrosis.The three HCl groups were intratracheally injected with HCl once per week.The control group was given saline by the same way.Six rats of each group were randomly sacrificed on day 7,14,28 and 42 respectively.The histological changes of lung tissue were studied by HE and Masson’s trichrome staining.Hydroxyproline level in lung tissue was measured by digestion method.Protein and mRNA expression of transforming growth factor-β1(TGF-β1) were assayed by immunohistochemistry and RT-PCR respectively.Results Alveolitis in three HCl groups was significantl compared to control group,most severe at the second week,then remained at a high level which was equivalent to or exceeded the level of the bleomysin group after 28 days.Pulmonary fibrosis in three HCl groups was also significantly more severe than that in the control group,but milder than that in the bleomysin group.The high-dose and middle-dose HCl groups were not significantly different from the bleomysin group on day 42.There was no difference between three HCl groups in the earlier period,but the high-dose HCl group has a significantly difference from low-dose group on day 42.The content of hydroxyproline in high-dose and middle-dose HCl groups was also significantly higher than that in the control group.On day 42 hydroxyproline content in high-dose HCl dose rather middle –or low dose group was similiar with the level of bleomysin group.Content of TGF-β1 mRNA in three HCl groups was comparable to the level of bleomysin group on day 28 and exceeded on day 42.The expression of TGF-β1 in three HCl groups was not significantly different from the bleomysin group on day 42.Conclusion Experimental acid aspiration might contribute to pulmonary fibrosis in rats.Acid induced alveolar epithelial cell damage,abnormal proliferation and repair and fibrosis could be involved..

  • Clinical Features of Microscopic Polyangiitis with Pulmonary Involvement in Comparison with Idiopathic Pulmonary Fibrosis

    Objective To explore the clinical features of microscopic polyangiitis ( MPA )complicated with pulmonary involvement in comparison with idiopathic pulmonary fibrosis ( IPF) . Methods Clinical and laboratory data of 27 patients with MPA and 56 patients with IPF in the Drum Tower Hospital from2006 to 2010 were analyzed retrospectively. The differences were compared between the MPA patients with pulmonary fibrosis manifestation ( MPA/PF patients) and those without pulmonary fibrosis manifestation( MPA/NPF patients) , and the IPF patients. Results The differences between the MPA/PF patients and the MPA/NPF patients were rarely found in terms of respiratory symptoms, ANCA positive rate, and multiple organ involvement, but the proportions of suffering severe renal damage and severe pulmonary hypertension in the MPA /PF patients were relatively high ( P lt; 0. 05) . Furthermore, there were significant differences between the MPA/PF patients and the IPF patients in terms of dyspnea, incidence of renal damage, ANCA positive rate, incidence of serious pulmonary hypertension, and multiple organ involvement. The IPF patients were more prone to develop dyspnea while MPA patients were more prone to develop renal damage, high ANCA positive rate, high incidence of serious PAH and multiple organ involvement, such as rush, joint pain,weight loss, fever and gastrointestinal symptoms ( P lt;0. 05) . Conclusions When patients have respiratory symptoms complicated with renal failure, skin damage, fever, and joint pain, the diagnosis of MPA should be considered. For patients who were clinically suspected as interstitial pneumonitis or pulmonary fibrosis,measurement of serumantineutrophil cytoplasmic antibodies and creatinine test are essential for diagnosis.

  • Immunoliposomes Containing Vasohibin Inhibit Pulmonary Fibrosis in Mice

    Objective To investigate the effects of immunoliposomes containing vasohibin on pulmonary fibrosis in mice. Methods Liposomes containing vasohibin plasmids were prepared by reverse phase evaporation technique, then combined with VWF antibody to form the immunoliposomes. 30 mice were randomly divided into a control group, a model group, and a vasohibin group. The mice in the model group and the vasohibin group were nasally instilled with bleomysin to induce pulmonary fibrosis. After 3 days, the mice in the vasohibin group were nasally instilled with the immunoliposomes containing vasohibin. The mice were all sacrificed after 14 days. Lung tissue sections were stained by HE and CD31 immunohistochemistry staining. The level of hydroxyproline in lung was measured by colorimetry. Results The resultsdemonstrated that the vasohibin immunoliposome could markedly decrease angiogenesis with the number of ( 46 ±16) compared to ( 78 ±12 ) , and decrease fibroblast proliferation with ( 0. 84 ±0. 12) μg/mg compared to ( 1. 39 ±0. 23) μg/mg. Conclusion The vasohibin immunoliposome can attenuate pulmonaryfibrosis and provide a novel strategy for clinical application in the future.

  • Effects of sodium ferulate on lung mRNA expression of TGF-β1 signal transduction molecule in pulmonary fibrosis rats

    Objective To investigate the effects of sodium ferulate on lung mRNA expression of TGF-β1 signal transduction molecule in rats with pulmonary fibrosis,and explore the mechanism of sodium ferulate on pulmonary fibrosis.Methods A rat model of pulmonary fibrosis was induced by intratracheal injection of bleomycin (5 mg/kg).Thirty SD rats were randomly divided into three groups (n=10 in each group),ie.a control group,a pulmonary fibrosis model group,and a sodium ferulate group.The lung histopathology and the expression of collagen was examined by HE staining and collagen fibril staining respectively.The expressions of TGF-βRII and Smad4 mRNA in the lung tissue were detected by situ hybridization.And the expression of TGF-β1 mRNA was detected by real-time fluorescence-quantification RT-PCR.Results Collagen fibril staining indicated that the expression of pulmonary collagen in the model group was significantly higher than that in the control group and sodium ferulate group (Plt;0.001).The mRNA expressions of pulmonary TGF-β1,TGF-βRII and Smad4 were significantly higher in the model group than those in the control group (all Plt;0.01),and were significantly lower in the sodium ferulate group than those in the model group (all Plt;0.05).Conclusions Sodium ferulate can effectively reduce pulmonary fibrosis through inhibition of the mRNA expression of TGF-β1,TGF-βRII and Smad4 in the lung tissue,thus influence the TGF-β1/Smad4 signal transduction way and inhibit the target gene activation.

  • Bone Marrow Mesenchymal Stem Cells Transform to Alveolar Epithelial Cells in Bleomycin Induced Lung Injury

    Objective To explore the migration and differentiation of bone marrow mesenchymal stem cells(MSCs) in lung . Methods MSCs were harvested from a male Wister rat. Sixty female Wister rats were randomly divided into four groups. The pulmonary fibrosis model was established by intratracheal instillation of bleomycin in group A-D. Immediately and 7 days after bleomycin administration respectively,the rats in group B and C received infusion with 5-bromodeoxynridine (BrdU) labeled MSCs via tail vein. And the rats in group D were infused MSCs without BrdU labeling serving as a negative control. The sry gene of Y chromosome was detected by polymerase chain reaction (PCR). Double immunofluorescence staining was used to detected BrdU and surfactant associated protein-C (SP-C) expression in lung tissue,fresh bone marrow,and the 5th generation MSCs. Reverse transcriptipon-PCR was used to detect the expressions of SP-C mRNA and AQP-5 mRNA. Results The sry gene was detected in bleomycin induced lung injury tissues of the rats after MSCs infusion immediately and on the 7th day The MSCs in lung tissue could transformed into cells with ACEⅡ morphological features and molecular phenotype. The transformation rate was higher in the rats received MSCs infusion immediately than the rats received on 7th day. The 5th generation MSCs and fresh bone marrow expressed SP-C mRNA,without AQP-5 mRNA and SP-C expression. Conclusions Exogenous MSCs can be transplanted into injured lung tissues and transform into AECⅡ,especially in early stage of lung injury. The differentiation potential of MSCs can be activated in injury micro-environment.
